Abstract : Recent advances in single-molecule force spectroscopy of DNA make it possible to study the thermodynamics and kinetics of DNA binding proteins under a wide range of conditions. A biophysical model for the DNA binding T4 gene 32 protein has been developed to study the kinetics of DNA protein binding to transient single-stranded DNA regions due to thermal fluctuations. The model is used to analyze recent single-molecule spectroscopy data of this system.
